We describe a new polarographic method to measure the haemoglobin oxygen saturation in whole blood, employing up to 10 μl of sample in a standard case. The measurement is done in an anaerobic staineless-steel cuvette (1 ml) recording three oxygen tension values: (1) that of an air-equilibrated buffer before the addition of the sample; (ii) that after the addition of the sample; and (iii) that after the addition of an oxidant. The haemoglobin oxygen saturation is then calculated from the three oxygen tension values, the volume of the reagents, and solubility coefficient of oxygen. This method is simple, inexpensive and accurate, and correlates well with other standard methods.  相似文献

在辽宁沈阳农田生态系统国家野外科学观测研究站,利用运行2a的开顶式气室,研究了臭氧(O3)浓度升高和不同氮肥施用水平对土壤线虫群落的影响。结果表明:(1)O3浓度升高降低了成熟期小麦根生物量。O3浓度升高和不同氮肥施用水平的交互作用改变了小麦成熟期土壤微生物生物量碳、氮和水溶性有机碳的含量。低氮条件下,O3浓度升高降低了土壤微生物生物量碳、氮和水溶性有机碳的含量;而高氮条件下则表现出相反的趋势。(2)O3浓度升高和不同氮肥施用水平对土壤线虫总数没有产生显著影响,而在灌浆期,食细菌线虫和食真菌线虫中c-p值为4(Ba4 and Fu4)的功能团对O3浓度升高和不同氮肥施用水平的响应敏感;与对照相比,不同氮处理中,O3浓度升高均降低了灌浆期Ba4功能团线虫的数量。灌浆期,O3浓度升高条件下,与对照相比Fu4功能团线虫数量在高氮条件下表现出增加的趋势,而在低氮条件表现出降低的趋势。(3)O3浓度升高和不同氮肥施用水平的交互作用显著影响了小麦灌浆期线虫的成熟度指数(MI)和结构指数(SI)。与对照相比,线虫成熟度指数和结构指数在低氮条件下随O3浓度升高而降低;而在高氮条件下随O3浓度升高而升高。上述结果表明,氮肥的施用能够缓解O3浓度升高对土壤食物网的扰动。  相似文献

Indoxyl glycosides proved to be valuable and versatile tools for monitoring glycosidase activities. Indoxyls are released by enzymatic hydrolysis and are rapidly oxidized, for example by atmospheric oxygen, to indigo type dyes. This reaction enables fast and easy screening in vivo without isolation or purification of enzymes, as well as rapid tests on agar plates or in solution (e.g., blue-white screening, micro-wells) and is used in biochemistry, histochemistry, bacteriology and molecular biology. Unfortunately the synthesis of such substrates proved to be difficult, due to various side reactions and the low reactivity of the indoxyl hydroxyl function. Especially for glucose type structures low yields were observed. Our novel approach employs indoxylic acid ester as key intermediates. Indoxylic acid esters with varied substitution patterns were prepared on scalable pathways. Phase transfer glycosylations with those acceptors and peracetylated glycosyl halides can be performed under common conditions in high yields. Ester cleavage and subsequent mild silver mediated glycosylation yields the peracetylated indoxyl glycosides in high yields. Finally deprotection is performed according to Zemplén.  相似文献

Background

Insertion sequences (ISs) are approximately 1 kbp long “jumping” genes found in prokaryotes. ISs encode the protein Transposase, which facilitates the excision and reinsertion of ISs in genomes, making these sequences a type of class I (“cut-and-paste”) Mobile Genetic Elements. ISs are proposed to be involved in the reductive evolution of symbiotic prokaryotes. Our previous sequencing of the genome of the cyanobacterium ‘Nostoc azollae’ 0708, living in a tight perpetual symbiotic association with a plant (the water fern Azolla), revealed the presence of an eroding genome, with a high number of insertion sequences (ISs) together with an unprecedented large proportion of pseudogenes. To investigate the role of ISs in the reductive evolution of ‘Nostoc azollae’ 0708, and potentially in the formation of pseudogenes, a bioinformatic investigation of the IS identities and positions in 47 cyanobacterial genomes was conducted. To widen the scope, the IS contents were analysed qualitatively and quantitatively in 20 other genomes representing both free-living and symbiotic bacteria.

Results

Insertion Sequences were not randomly distributed in the bacterial genomes and were found to transpose short distances from their original location (“local hopping”) and pseudogenes were enriched in the vicinity of IS elements. In general, symbiotic organisms showed higher densities of IS elements and pseudogenes than non-symbiotic bacteria. A total of 1108 distinct repeated sequences over 500 bp were identified in the 67 genomes investigated. In the genome of ‘Nostoc azollae’ 0708, IS elements were apparent at 970 locations (14.3%), with 428 being full-length. Morphologically complex cyanobacteria with large genomes showed higher frequencies of IS elements, irrespective of life style.

Conclusions

The apparent co-location of IS elements and pseudogenes found in prokaryotic genomes implies earlier IS transpositions into genes. As transpositions tend to be local rather than genome wide this likely explains the proximity between IS elements and pseudogenes. These findings suggest that ISs facilitate the reductive evolution in for instance in the symbiotic cyanobacterium ‘Nostoc azollae’ 0708 and in other obligate prokaryotic symbionts.

目的:研究精神疲劳状态下脑组织血氧饱和度的变化规律。方法:从某军校随机抽取25名被试,采用模拟飞行任务负荷构建精神疲劳模型,采用NASA-TLX量表评价模型。实验全程使用近红外光谱技术对被试脑组织血氧饱和度进行实时监测;实验前后分别对被试的作业绩效水平进行评估。结果:任务后NASA-TLX量表评分明显高于任务前(P0.01);任务前后作业绩效发生变化,反应能力测试的正确率下降(P0.01),错误率上升(P0.01);任务负荷后脑组织血氧饱和度相对于静息状态升高(P0.05)。结论:精神疲劳状态会影响被试的作业绩效。疲劳后脑组织血氧饱和度水平受被试动机以及代偿机制的影响高于静息水平。  相似文献

【目的】采用Plackett-Burman Design (PBD)与均匀设计(Uniform Design, UD) 优化玫瑰色微球菌(Micrococcus roseus)固定化脱氮的条件,以提高其脱氮性能。【方法】选择聚乙烯醇(Polyvinyl alcohol,PVA)与海藻酸钠(Sodium alginate,SA)作为固定化细胞材料,研究其成球方式与性能,应用PBD对9个影响脱氮效果的因素进行显著性检验,筛选出PVA、SA、温度、菌液接种量、菌球量5个显著性因素(P<0.05)。采用UD对5个显著因素进行条件优化,优化数据通过Minitab与Matlab软件进行逐步回归分析,并建立二阶多项式模型。【结果】最优脱氮条件为PVA 9.6%、SA 2.2%、温度33.4 °C、菌液接种量8%、菌球量500个/100 mL,连续脱氮72 h,最高脱氮率达到60.9%。【结论】PBD与UD的联合使用能够在显著因子筛选与寻优中发挥重要的作用。连续脱氮试验表明,玫瑰色微球菌去除氨氮主要通过同化、硝化与反硝化途径。  相似文献

The purpose of this study is to investigate the nitrogen removal performance of the anaerobic ammonium oxidation (Anammox) process and the microbial community that enables the Anammox system to function well at ambient temperatures. A reactor with a novel spiral structure was used as the gas-solid separator. The reactor was fed with synthetic inorganic wastewater composed mainly of NH4+-N and NO2-N, and operated for 92 days. Stable nitrogen removal rates (NRR) of 16.3 and 17.5 kg-N m−3 d−1 were obtained at operating temperatures of 33 ± 1 and 23 ± 2 °C, respectively. To our knowledge, such a high NRR at ambient temperatures has not been reported previously. In addition, the experiments presented herein confirm that high influent NO2-N concentration of 460 mg L−1 did not noticeably inhibit the Anammox activity. Furthermore, the freshwater Anammox bacterium KU2, which was identified as the dominant bacterial species in the consortium by 16S rRNA gene analysis, is considered to be responsible for the stable nitrogen removal performance at ambient temperatures.  相似文献
